چکیده
demonstrate that molecular hydrogen but not molecular nitrogen inhibits the splitting of H2 from pyruvate by Clostridium pasteurianum. The effect of H2 is similar to that observed by Kubowitz (Biochem. Z., 274, 285, 1934) with Clostridium butyricum and probably merely reflects a shift in the equilibrium of the reaction. The findings were not affected by substituting argon for the helium. From the point of view of comparative biochemistry, it is of interest that Rhodospirillum rubrum differs from Clostridium pasteurianum not only in the absence of a "Kamen-Gest reaction" in the latter but also in the absence of an effect of H2 on the photoevolution of hydrogen by the photosynthetic anaerobe. The organisms are similar in that H2 does not appear to inhibit nitrogen fixation by either.

Periplasmic expression of Bacillus thermocatenulatus lipase in Escherichia coli in presence of different signal sequences
Efforts to express lipase in the periplasmic space of Escherichia coli have so far been unsuccessful andmost of the expressed recombinant lipases accumulate in the insoluble cell fraction. To evaluate the role ofnative and heterologous signal peptides in translocation of the lipase across the inner membrane of E. coli, the lipase gene (btl2) was cloned downstream of the native ...
متن کاملOBSERVATIONS ON THE CHROMATINIC BODIES OF Streptomyces coelicolor
Colonies of Streptomyces coelicolor growing on cellophane and impression preparations from sporing colonies were stained for chromatin by the methods of Feulgen, DeLamater (1951), and Piéchaud (1954). The chromatinic bodies of the substrate hyphae have a great variety of configurations. During the development of the spores, elongated chromatinic structures in the young aerial hyphae separate in...
متن کاملEffects of Protoplast Fusion on δ-endotoxin Production in Bacillus thuringiensis Spp. (H14)
In this study, mutant forms of Bacillus thuringiensis spp. israelensis (H14) were produced. These mutants were identified when the cells were cultured on chloramphenicol plates and stained with crystal violet. Protoplasts of the mutants were isolated by enzymatic digestion (lysozyme) of the cell walls at the presence of an osmotic stabilizer. The protoplasts were induced to fuse to each other i...
متن کاملGenome-Scale Metabolic Network Models of Bacillus Species Suggest that Model Improvement is Necessary for Biotechnological Applications
Background: A genome-scale metabolic network model (GEM) is a mathematical representation of an organism’s metabolism. Today, GEMs are popular tools for computationally simulating the biotechnological processes and for predicting biochemical properties of (engineered) strains. Objectives: In the present study, we have evaluated the predictive power of two ...
متن کاملMOLECULAR CLONING AND EVALUATION OF WILD PROMOTER IN EXPRESSION OF BACILLUS SPHAERICUS PHENYLALANINE DEHYDROGENASE GENE IN BACILLUS SUBTILIS CELLS
To evaluate the role of wild promoter of L-phenylalanine dehydrogenase (PheDH) gene, referred to as pdh, from Bacillus sphaericus in expression, cloning of pdh gene in Bacillus subtilis was performed. The whole pdh gene was cloned in pHY300PLK shuttle vector and amplified, construct (pHYDH) then transformed in B. subtilis ISW1214 and E. coli JM109. The pdh endogenous promoter presented no effec...
